Processing a ROI might result in a different noise filter as compared to processing the whole image
dependent on the image content.
If a Z-Stack was acquired you have the option to
tell the system, which planes of the stack should
be processed from the
Stack processing mode
drop down box (Fig. 375).
You have the option between
All Z
,
First-Last
or
Current Z
.
If
All Z
is selected, the whole stack will be
processed.
When
First-Last
is selected the double-slider or the left and right spin input boxes can be used to define
the first and last frame of the stack that should be processed (Fig. 376).
If
Current Z
in
SIM
is selected, the current frame
selected in the
2D view
will be processed.
In
SIM
processing tool you have the Option for fast
Preview Processing by pressing the
Preview
Processing
button (Fig. 377). The systems selects a
ROI in XYZ by means of highest contrast in Z and
highest intensity in XY and calculates the fitting
parameters if in the
Adjust
window in the
Advanced Filters
field the
Parameters
drop
down menu was set to
Fast
.
Note, if
Best
is selected the whole image will be taken and computation will be accordingly slower. If a
ROI is defined and the Use ROI XY checkbox ticked the system will display the SIM image for this ROI
only. Moving the ROI will update the SIM image on the fly. This might help to fast evaluate the fitted
parameters and make adjustments for the processing of the total image. As previewed parameters will be
taken for computation of the whole image, processing is much faster than processing an image without
Preview Processing.
If more channels have been acquired the
Settings
for Channel
drop down menu allows you to set
parameters channel specific (Fig. 378). You have
the option to select
All
or single channels from the
multi-channel image that should be processed. If
All is selected, the current settings will be applied
to all channels. If one channel is selected, the
settings defined for each channel will be applied to
each of them. If a The
Settings for Channel
drop
down menu is not greyed if the loaded image has
only one channel.
If the
Sharpness Auto
box in
SIM
is not ticked,
the
Sharpness
filter slider is available, that can be
used to set the noise filter for the so called Wiener
de-convolution (Fig. 379). The number indicates
the negative number to the power to 10 for the
filter.
The smaller the values (towards -11), the higher
the frequencies that are allowed leading to noisier
and crispier images.
