(8) The Definite Focus tool during
synchronous stabilization
Whenever the
Definite Focus
is selected from the
Autofocus Mode
drop down menu of the
Focus
Device and Strategy
tool (Fig. 168) as the focus
strategy, this setting will overrule the settings made
in the
Definite Focus
tool whenever a scan is
triggered by
Start Experiment
.
When Definite Focus is selected from the
Autofocus
Mode
drop down menu, the
Autofocus every n
Timepoint / Position / Tilescan
check boxes with
their input boxes become available, dependent on
the chosen dimensionality of the experiment.
To run in synchronous mode please check the boxes
accordingly and enter an appropriate number in the
input box defining before which events a stabilization
should occur (Fig. 169).
Stabilization before each time point (enter
1
in
Autofocus every n Timepoint
) is the most
frequent stabilization scheme possible.
No continuous stabilization can be triggered from within the
Focus Device and Strategy
tool.
Stabilization from the
Focus Devices and Strategy
tool runs synchronously to the scan. In
contrast, stabilization from the
Definite Focus
tool will be asynchronous.
The state of the
Use Z Piezo
check box is applied for the stabilization via the
Focus Devices and
Strategy
. That means if the box is checked, the Z-Piezo will be used.
With
Definite Focus
selected in
Focus Devices and
Strategy
tool the
Find Surface
and
Periodic
buttons will be deactivated during a scan in the
Definite Focus
tool (Fig. 170), no matter if the scan
was triggered by
Continuous
or
Start Experiment
.
If
Periodic
is active and the
Continuous
scan button
pressed, the stabilization, even if the
Periodic
button
becomes inaccessible, will run with the respective
settings in the background.
When the
Stop
scan button is pressed, the once
active
Periodic
button becomes active again.
Stabilization will end only if the active button is
pressed again. In contrast, if
Start Experiment
is
pressed the
Periodic
button becomes inaccessible as
well, but the stabilization regime is executed as
defined under
Focus Devices and Strategy
.
