Collimator Setting
The Collimator setting is specific for the
Laser WF
imaging mode.
•
Press the
Collimator
button to open the
Collimator settings
panel (Fig. 101).
•
Adjust the Collimator position using the
Collimator Camera ELYRA
slider or by typing
into the input box or using the arrows. The
value in the input box and below the Collimator
button display the current motor step position
(
Pos.
).
•
By pressing the
Save collimator position(s)
button, the current position is saved. You will
be prompted in the
Collimator
window to
confirm (Fig. 102). Press
Yes
to save the new
position,
No
to abort the saving process. By
pressing the
Reset
button (only active once a
Collimator position was saved), the stored
collimator position will be recalled.

TIRF Angle Setting
Please be aware that for systems without
3D-PALM slider, you have for the
TIRF angle
settings panel
EPI
and
TIRF
mode available.
In systems with 3D-PALM slider you have
EPI
,
HILO
and
TIRF
mode as options.
TIRF angle
setting is specific for the
Laser WF
imaging mode. Press the
TIRF angle
button to
open the
TIRF angle settings
panel (Fig. 103).
You can switch between
EPI, HILO
and
TIRF
mode
by pressing the respective button. In
TIRF
mode, a
slider becomes available, with which one can set
the TIRF angle.
The HILO angle is arbitrarily offset from the TIRF angle by 10 mechanical steps of the TIRF mirror.
You can also input a value in the input box or use the arrows to change the value. The value displayed in
the input box and above the
TIRF angle
button display the
Angle
in degrees (
0
). The green outline
defines the region with ±1
0
from the stored centered position.
By pressing the
Save TIRF angle
button you will be prompted to confirm saving the current position in
the
TIRF Mirror
window (Fig. 104). Press
Yes
to save the new position,
No
to abort the saving process.
By pressing the
Reset
button (only active once a Collimator position was saved), the stored collimator
position will be recalled. The theoretical calculated
Penetration
depth for each laser line is given in nm in
the
Penetration
field.
