If in the
Laser WF
imaging mode at least
one laser is selected, the
Laser Trigger
expansion field becomes available (see
Fig. 141).
When expanding the box, all active lasers
will be displayed with their
Trigger
drop
down menus. You can select between
−
none
: no trigger
−
integration
: the laser is on only
during the integration of the camera
−
transfer
: the laser is only on during
the frame transfer of he camera
Note that the laser trigger only works in the
Laser WF
imaging mode for the
Andor iXon SMLM
and the
CMOS pco.edge SIM
cameras.
For the
SIM Apotome
,
Lattice
SIM
and
SMLM
imaging modes the Field of View (
FOV
) expansion field
is available (see Fig. 142). By opening the field you can select the field of view via the
FOV
drop down
menu (see Fig. 142). You have the following options
−
SIM
for the SIM imaging mode
−
TIRF
for the laser WF imaging mode
−
HP
(high power field)
for the laser WF imaging mode
−
uHP
(ultra high power field) for the laser WF imaging mode
In the
Laser WF
under
Illumination
mode
you can select the illumination scheme you
want to use. By pressing either of the
buttons
EPI
(Epifluorescence),
HILO
(Highly
Inclined Laminar Optical Sheet) or
TIRF
(Total Internal Reflection Fluorescence). The
active button is highlighted (see Fig. 142).
You can also press the
3D-PALM
button to
activate the 3D-PALM slider to move into
the back aperture of the objective once an
active Scan is started.
The button is highlighted, if active. Press the button again to deactivate and to put the 3D-PALM slider in
its rest position. When active online processing will be disabled.
By pressing the
Save TIRF angle
button the current set TIRF angle will be saved that will be recalled
when pressing the
Reset
button on it right. By pressing the
Save Collimator position(s)
button the
current set collimator position will be saved that will be recalled when pressing the
Reset
button on its
right.
The
Channel
display field shows all selected channels with their name. Each channel can be controlled
separately. In the
WF
,
SIM
and
Laser WF
an EMCCD camera serves as the detector for each channel.
You can set the exposure time by and the EMCCD gain via the
Exposure Time [ms]
and
EMCCD Gain
sliders, respectively or their input boxes.
The
Camera Temperature
display field shows the current status of the camera’s temperature.
