Acquisition Parameter – Acquisition
Mode
In the
Acquisition Mode
tool window,
parameters for the acquisition can be set
(Fig. 111,).
The user interface is adapted depending on the
chosen Imaging mode in the
Imaging Setup
tool.
Controls include

Objective selection

Frame Size selection
−
Format selection
−
Averaging and color depth selection
−
HDR High Dynamic Range (not available)
−
Scan Area selection (not available)

Objective Selection
You can select an objective from the
Objective
drop down menu (Fig. 112). Objectives suited for a
certain imaging mode will be highlighted, but
every objective can be selected.
In case the system is equipped with an objective
that has a motorized correction ring, an additional
slider appears beneath the
Objective
selection
(
Corr (%)
) once this objective is selected.
The slider is used to set the correction ring to the
best position for minimal spherical aberration
(Fig. 113).
Ideally use a line z scan (time series with maximum
number of time points) to look at the reflection of
the coverslip. Set the slider to a position such that the image shows the reflection of the line to be as thin
as possible. This is the optimal correction for minimum spherical aberration.
Default
sets the correction
back to its middle position.
Note that the default value set by the
Default
button is not always the middle of the correction
collar, but the point where the optical design works at its theoretical best performance.
