If
Apply
is pressed “Line-scan” plots (and the corresponding table) will be displayed in the image
container with which the modulation contrast can be assessed (Fig. 347).
The result is showing the dependence of the
Intensity
of the 1
st
and 2
nd
orders on z (
Distance [
µ
m]
). The
channel code
TV#-TX#-RX#-O#
is as follows:
−
TV#-T# = channel name
−
R# = number of rotation
−
O# = number of order
The 1
st
orders should show periodicity (due to the Talbot effect), whereas the 2
nd
and higher orders should
be more or less constant over the whole z-range. As an output there will also be two files in the
C:\\Temp
folder called
ModulationContrast.txt
and
ModulationPeriod.txt
.
ModulationPeriod.txt
contains the grating period for different rotations and z-planes as estimated from the modulation curves.
This estimate is independent of the main SIM processing and can be used for cross-check.
ModulationContrast.txt
contains information for every localized bead: position, intensities and phase in
the first and second orders. This file can be used for further possible quantitative analysis of the
modulation.
