Switching on
•
Turn on the entire system via the three
switchable distributor sockets.
•
Swivel the objective into the beam path (use a
filter that is suitable for the desired laser line).
If the Slider Laser TIRF has not yet
been placed in the receptacle of the
luminous-field diaphragm, insert it
according to Section 3.9 (see
page 70).
•
In the case of the Slider Laser TIRF man., rotate
the TIRF angle adjusting screw (Fig. 4-1/
7
)
carefully to mid-position.
•
Put the specimen on the stage and close the
laser safety cover or the laser safety incubator.
•
Turn on the laser (key switch on Laser Bench
Tower or Rack to
ON
).
•
Switch on the ON / OFF switch (Fig. 4-4/
1
) of
the signal distribution box. The green pilot LED
integrated in the switch lights up.
•
Switch on the ON / OFF switch (Fig. 4-5/
4
) of
the TIRF controller.
The four pilot LEDs (Fig. 4-5/
1
,
2
,
3
,
5
) light up.
The red and the yellow pilot LEDs (Fig. 4-5/
2
and
3
) will go out after about 5 seconds.
If the red and the yellow pilot LEDs do not go out after 5 seconds, a system error has
occurred. Please contact the Carl Zeiss Microscopy GmbH Service department.
•
Start the ZEN software (blue edition) when using Slider Laser TIRF mot. In this case the Slider Laser TIRF
is controlled by the software.
The device is ready for use when the
Active
and
Safe
LEDs are lit. See Sections 4.4 to 4.7.
If the safety system does not work in the way described above, take the instrument out of use,
label it clearly as being out of order and immediately contact Carl Zeiss Microscopy GmbH
Service.
Before working on the actual sample, the system should be checked to ensure that the laser beam is
correctly focused, see Section 4.5.2 on page 77.
