Troubleshooting
TIRF microscopy requires aqueous specimens! Please note that the evanescent field is
approximately 200 nm thick. Only specimens within this field can be observed through the
microscope. Focus the specimen correctly. Take care that there is no dust on the objective or in
the oil.

No laser light
Laser safety cover or incubator not correctly
placed.
Place laser safety cover or incubator
correctly.
Slider Laser TIRF not inserted.
Insert Slider Laser TIRF, see Section
3.9 on page 70.
Connections to distribution box not
established.
Connect the Distribution Box for Laser
Safety
Transmitted light illumination carrier tilted
backwards.
Bring transmitted light illumination
carrier to vertical position.
Reflector turret not clicked into position or
pulled out.
Click reflector turret into position or
push it in.
No TIRF effect
Epi fluorescence illumination is switched on
Switch off epi-fluorescence
illumination by pressing the
RL
button.
Object to be examined is outside the
evanescent field (approx. 200 nm thick).
Look for an object situated on the
cover slip and focus it.
Wrong reflector module selected.
Select appropriate reflector module
(according to wavelength).
Specimen is embedded in resin or a similar
substance so that critical angle for total
reflection cannot be reached.
TIRF microscopy requires aqueous
specimens!
Do not remove the connections from the Laser Bench Tower or Rack under any circumstances.
The Laser Bench Tower and Rack may only be connected by Carl Zeiss Microscopy GmbH
Service staff.

Check that the connections on the Laser Bench Tower and Rack are tight.
