determined by depth of focus of the objective. A higher resolution uses a smaller z
increment.
The estimated time to collect is displayed in the Terrain Mosaic Setup dialog. The
collection time depends on the resolution, total z-range, and collection settings.
20. Click
Start
.
The instrument begins collecting a terrain mosaic. Click STOP Mosaic to stop collecting
the terrain mosaic before the mosaic is complete.
When the terrain mosaic is collected, the terrain map opens automatically.
21. In the Terrain window, use the slider to adjust the
smooth strength
. The smoothing
affects how spectral data is collected. Use the image of the terrain map to find a balance
that reduces noise while preserving sufficient surface definition. Noisy data may look like
fur, with many high and low points. If smoothing is too strong, you may loose important
details in the sample surface.
22. When you are satisfied with the smooth strength, close the window. Smooth strength is
saved automatically.
You can now use the terrain mosaic to collect a chemical region or to analyze particles.
To use the terrain information when collecting a region or analyzing particles, open the
Region Queue tool to ensure that Terrain is selected in the Terrain column. Clear the
selection to ignore the terrain mosaic and to instead use a single z position during
collection.
Acquire and Analyze Data
Use the OMNICxi software to do the following:
•
: Collect a chemical image of one or more regions of the sample surface.
Specify regions automatically or select only specific areas of interest with manual regions.
•
Identify and Analyze Particles
: Use the particle selection tools to locate and characterize
particles in the sample.
•
View Spectrum at Specific Location
: Select Live Spectrum mode to view the spectrum of
a specific location on the sample surface.
•
: With Image Cross Section mode, collect a cross section of
transparent sample.
•
: Acquire a series of measurements to analyze how your
sample changes over time.
•
: View 3D data sets in the 3D volume viewer.
