The instrument should be powered up and running for at least one hour before you align and
calibrate a laser set. OMNICxi software should be running and the status indicator in the
upper right corner should show a green check mark. Wait at least 5 minutes after changing the
installed grating before you align and calibrate a laser set.
Tools needed:
• Align/calibrate tool (normally stored in the holder attached to the left side wall inside the
microscope sample compartment and left connected to the instrument)
To align and calibrate the current laser, grating and filter set
1. Open the sample compartment doors.
2. Make sure the
standard (single slide) sample holder
is installed on the microscope stage.
3.
Install the alignment and calibration tool
on the single slide sample holder.
Make sure the tool is in the detent, level and secured by the stage clip and both
connectors are secured.
4. Rotate the 10x objective into the front objective position so it engages the detent. (Not
necessary to select the objective in OMNICxi software for this procedure.)
5. From the OMNICxi Acquisition window, click
Settings
and then click
.
A message appears.
6. Use the stage controller to move the stage so that the alignment tool is under the white
light from the illuminator.
Note
If the currently installed laser set does not have associated alignment and calibration
data, the status indicator will show a red “X.” If you see a red X that indicates the current
laser set is uncalibrated, follow the steps below to create alignment and calibration data for
this laser, grating, and filter set.
