Image Processing
Use the Image Processing group to specify how the selection
handles particles with holes, touching particles, and partially
visible particles.
The following tools are available:
•
Fill Holes Whose Size Is.
If detecting particles as a
region, data from the hole will be ignored. Use the slider
to set a threshold for how large of a hole should be
ignored.
•
Separate Touching Particles.
Select to treat particles
that are touching as if they were separate particles.
Clear to treat the group as a single particle.
•
Exclude Partially Visible Particles.
Excludes partial or
incomplete particles that are cut off on the edge of the
mosaic or region.
Background Intensity
Use the background intensity group to limit the selection
based on visual contrast.
The following tools are available:
•
Auto Detect.
Automatically detects what is a
background and what is a particle based on visible
contrast. The histogram shows what percentage of the
image is a given intensity. Intensity is a measurement of
how light or dark a pixel is, from completely black to
completely white, with white being the most intense. If
you have a very light background with only dark
particles, the histogram may show that more than 80%
of the image is high intensity.
Clear to refine the selection with the image intensity
histogram.
•
Use 2 regions.
Select to manually refine the selection
using to regions on the histogram. This is useful for
selecting particles that are lighter and darker than a
neutral background.
•
Image Intensity Histogram.
Manually refine the
selection based on visual contrast.
