14. With the Terrain Mosaic Setup open, use the preview pane and the stage controls to bring
the highest part of the sample into focus. This will be the lowest stage position with the
sample farthest from the objective. When you have found a high point on the sample,
click
Focus Video
to bring the sample into focus.
15. With the highest part of the sample surface in focus, next to Z minimum, click
Use
Current Z
to set the Z minimum. This will be the lowest stage position used to collect
the terrain mosaic.
16. Using the preview pane and stage controls, move the stage toward the lens to bring the
lowest part of the sample into focus. This will be the highest stage position with the
sample nearest to the lens. Click
Focus Video
to bring the sample into focus.
17. With a low point on the sample surface in focus, next to Z maximum, click
Use Current
Z
to set the Z maximum. This will be the highest stage position used to collect the terrain
mosaic.
18. Select an overshoot percentage. The overshoot adds a percent of the total z range to
maximum and minimum z position to ensure that any points just outside the z range are
in focus.
The overshoot percent is added to both maximum and minimum. For example, with a
full range of 300 μm, 30 μm will be added to both the max and min, for a total z range of
approximately 360 μm. The total z-range is rounded to fit the z increment, so the range
may be somewhat larger than the exact percent overshoot.
An overshoot percent of 5% is good for most applications. You may need a larger range if
your sample is very rough or uneven. Use 0% if you are concerned about bumping the
sample into the lens.
19. Select high, medium, or low z-step resolution. A high resolution takes longer to collect,
but collects more z positions, resulting in more accurate data. The z-step resolution is

When adjusting the stage position, be careful not to bump the sample into the
microscope objective. The sample will be very close to objective at the highest stage
position. Use a long working distance lens allow more space between the sample and the
lens.
