CEQ™ 8000 Genetic Analysis System
Primer Label (optional)
The
Primer Label
area of the window allows you to indicate (for supplementary
reporting purposes) on which strand the primer is labeled with the dye.
•
Select the
forward
option button if the primer is dye labeled on the forward
strand.
•
Select the
reverse
option button if the primer is dye labeled on the reverse strand.
•
Select the
none
option button if you do not want to indicate which strand the
primer is dye labeled.
Primer Sequence (optional)
The
Primer Sequence
area of the window allows you to indicate (for supplementary
reporting purposes) the specific sequence of the primer used. This can be up to 128
characters.
•
If the primer is labeled on the
forward strand
, input the sequence of the primer
in the
5' to 3' direction
.
•
If the primer is labeled on the
reverse strand
, input the sequence of the primer
in the
5' to 3' direction
.
Generating and Viewing the Allele List
Once you have made your selections, you may choose to begin creating the allele list
by clicking the
Generate Allele List
button.
The system generates an allele list from the information provided by the user. The
allele list starts at the low end of the fragment length range and creates an allele at each
perfect repeat length until the end of the fragment length range is reached.
The generated fragment lengths are displayed in the
Nominal Size
column as the
initial integer values. These values can be user-modified to represent the allele sizes
that appear when the data is analyzed. The nominal sizes are also displayed in the
apparent size column, but these will usually be edited or regenerated.
The
ID’s
assigned to each allele in the list use the allele naming convention described
in the section above.
The fragment lengths displayed in the
Apparent Size
column are either user-entered
from the locus tag editor, generated by interpolation or regression from an incomplete
list, or generated from the automatic binning analysis feature (see the “Bin Analysis”
section).
When apparent fragment lengths are user-entered or are generated by interpolation or
linear regression, the nominal sizes must have first been entered by the user. When the
apparent fragment lengths are generated by binning analysis, the nominal sizes are
automatically generated.
