3. Check operation of LED.
4. Create a Sample with an RBC count of 0.80±0.1M/µl by diluting a Low Control:
a. Run Low Control or sample to obtain RBC count.
b. Multiply RBC count by 1.25.
c. Subtract 1 from Step 4b.
d. Multiply Step 4c by Volume of sample.
e. Add volume of Diluent calculated in Step 4d to sample to get an RBC count of .8 +/- .1 M/µl.
Example
RBC Count = 2.75 M/µl
Sample volume used to create dilution = 500µl
Multiply count by 1.25: 2.75 x 1.25 = 3.438
Subtract 1.0 from result above: 3.438 - 1.0 = 2.438
Multiply result above by sample volume:
2.438 x 500µl = 1220µl =1.22ml
So, a 0.80 M/µ RBC is created by adding 1.22ml of diluent to 500µl of a sample with an RBC
