When the
Save Default
button is pressed, the
present coefficients will be stored and can be
recalled by pressing the
Restore default
button,
which becomes active once the
Save default
button was pressed once.
If the
Input 2
check box remains inactive, the image
selected for
Input image 2
is not considered for
alignment. If the
Input 2
check box is ticked, than the
channels of
Input image 2
are aligned to the
corresponding channels of the
Input image
that is to
those channels at corresponding positions in the list.
It is the responsibility of the user to align only images of
the same dimensions and channels of the same format. If
images will have different dimensions, for example a raw
SIM Z-Stack and a raw PALM time the system will give no
warning message telling the user that the images do not
match (Fig. 226). It will load both images and aligns the
channels according to their position as long as there are
matching channels.
If no
Input image
was selected and only an
Input
image 2
, no alignment will be performed.
Loaded images cannot be unloaded but replaced
by other images in selecting the respective image
in the image container and then pressing the
Select
button for
Input image
or
Input image 2
.
If the
Fit check
box is not ticked a second table, the
assignment table
will be displayed below the
coefficient table
(Fig. 227). In this case the selected
image will not be fitted directly, but a loaded alignment
will be performed on the image. Columns in the
coefficient
and
assignment
tables can be expanded by
dragging the borders of the columns.
The
coefficient table
in this case has to be loaded from
a .bin file by pressing the
Load
button or is still available
from a previous alignment or obtained by pressing the
Restore Default
button, if values have been previously
restored by pressing the
Save default
button. In any
case, the listed coefficients (no matter what
Alignment
mode
is associated) will be applied to the selected
image. If the
Input 2
check box is inactive, this will be
the image selected for
Input image
. If the
Input 2
check box will be ticked, it will be the image selected for
Input image 2
.
