Plastic Object Holder
The plastic Object Holder is made of material that does not conduct heat
effectively.
If the interior of the chamber cannot be cooled, using this Object Holder
keeps the tissue sample frozen for a longer time because it is less
affected by the temperature of the specimen head compared to the
standard Object Holder.
If the interior of the chamber can be cooled properly, use the
standard Object Holder.
For the plastic Object Holder, only the square type is available.
There is no round type.
Using the Plastic Object Holder
If the interior of the Polar® instrument chamber cannot be cooled for some reason and sectioning must be
performed urgently, use the method below to perform sectioning.
1. Remove the blade holder from the chamber and keep it in a deep freezer, etc., for at least 2 hours to cool
the holder.
Refer to p. 2-32 for information on how to remove and set the blade holder.
Cool not only the stage, but the blade holder as well.
2. Prepare frozen blocks.
For the Object Holder, use the plastic Object Holder that does not conduct heat effectively.
• Put tissue specimens in the Cryomold.
• Put OCT compound by making sure it does not overflow.
• Put the plastic Object Holder on top.
Make sure the side to be set on the specimen head (circular shape) faces up.
Freeze the specimens according to the "Liquid Nitrogen Method," "Dry Ice Acetone Method," "PINO
Hyfluid Method," etc.
3. When the blade holder has been cooled, set the blade holder in the chamber.
Refer to p. 2-32 for the setting method.
4. Next, set the frozen block on the specimen head.
Refer to p. 2-6 for the setting method.
If the specimen head cannot be cooled, cool it with a cooling spray beforehand.
5. Section the block.
Refer to p. 2-11 for the sectioning method.
Sectioning must be completed in approx. 50 to 60 seconds.
An effective way to extend the cooling time is to use a cooling spray to cool the specimen head and
blade holder while sectioning.
The Cryomold and Object Holder used should have appropriate shapes.
It is not guaranteed that tissue specimens similar to what can be expected from the standard Object
Holder can be prepared.
It is recommended that trial runs are performed beforehand by also assuming emergency scenarios.
