Pipette
Offset
is
used
at
the
beginning
of
each
experiment,
when
the
pipette
is
first
introduced
to
the
bath,
to
compensate
for
potentials
caused
by
differences
in
the
concentration
of
bath
and
pipette
solutions.
How
to
Use
Pipette
Offset
Introduce
the
pipette
to
the
bath
and
click
the
Zero
button.
A
test
pulse
is
generated,
potential
calculated,
and
offset
automatically
applied.
The
associated
padlock
button
“locks”
and
the
Pipette
Offset
button
is
disabled.
Manual
adjustments
can
be
made
to
the
setting
following
use
of
the
Zero
button.
Adjust
the
glider
control
until
the
reading
in
the
current
meter
is
zero.
The
allowed
range
is
±250
mV.
The
glider
control
allows
fine
adjustments
with
increments
of
20
μ
V
for
current
clamp.
To
reset
the
Pipette
Offset,
click
on
the
“locked”
padlock
button
to
“unlock”
it,
and
click
the
Zero
button
again.
Note
the
cautionary
point
below,
however.
Additional
Notes
Auto
Pipette
Offset
can
fail
if
the
DC
voltage
is
greater
than
±250
mV.
To
avoid
introducing
errors,
you
should
not
change
the
pipette
offset
after
penetrating
a
cell
with
a
sharp
electrode,
or
after
switching
from
voltage
‐
clamp
to
current
‐
clamp
mode.
Pulse
The
Pulse
command
is
located
near
the
bottom
of
both
V
‐
Clamp
and
I
‐
Clamp
tabs.
It
includes
an
activation
button,
and
adjustable
fields
for
pulse
amplitude
and
duration.
Pulse
provides
a
way
of
generating
simple
command
currents
(I
‐
Clamp)
and
command
voltages
(V
‐
Clamp).
The
amplitude
and
duration
of
these
can
be
set
in
the
associated
fields.
