Do not place the Gas Mixer and the Humidifying Column on the microscope stage! The vibration
might disturb the measurement.
After connecting all cables and tubing, and after filling the Humidifying Column with ultra-pure water, the
system is ready to start.
1.
Set the gas regulation valve to the recommended pressure of 1 bar (14.5 psi).
2. Switch on the Gas Mixer and wait until bubbling is observed.
3. Set the parameters in the ZEN Blue Lattice Lightsheet incubation tab (Fig. Fig. 26).
4. Give the system time to equilibrate all parameters for a minimum of 30 minutes.
Be aware that when the system is switched on, some parts of the tubing are actively heated:
−
the Humidifying Column base (max. 55°C/131°F)
−
the water in the Humidifying Column (max. 60°C/140°F)
−
the Heated Tubing Package (max. 40°C/104°F outside)
