Sample Imaging
Setup volume scan in
Acquisition Mode
(Fig. 67). Either use Center mode or First/Last
mode.
Set the beam path as required.
Center Mode:
1.
Press Center to center the stage at the current
position. Otherwise, the stage will do the scan
around its last position, which could be far off
(14 cm in worst case). If the Auto-Center
checkbox is ticked the system will center around
the current position of the stage automatically.
2.
Define the Scan range (
Range y
) or the number
of slices (
Slices
)
3.
Define the step size or (
Interval
).
Default interval is 0.2 µm for Nyquist-sampled
imaging. This should only be changed to
coarser values if experiment / data rate / speed
allows for.
4.
Limit acquisition to the minimal necessary
camera sensor area to reduce camera read out
time and data volume (optional)
5.
Set time series as required.
6.
Set environmental conditions as required.
7.
Press
Start experiment
to execute a single 3D
volume imaging or a time series as defined
(Fig. 68).
