Staining Performance

Optimal staining results depend on the quality of tissue and the way in which it is processed prior to staining.

Problem

Possible Causes & Solutions

No staining, weak or inconsistent
staining, false negatives�

Cause: Programming errors.
Solution: Review programming and correct errors.

Cause: Slides positioned incorrectly in slide racks. Instruments and slides must be
horizontal, verified by level gauge.
Resolution: Reposition slides.

Cause: Slide labels positioned incorrectly onto the slide. If the slide label is not properly
placed and sticks up even slightly, liquid flow will be impeded and reagents will wick
and yield poor staining quality. Please see the diagram in the Slide Preparation section
of chapter 5.
Resolution: Reposition slide labels.

Slides have dried out�

Cause: Slides positioned incorrectly on the VALENT
Resolution: Visually inspect the slides from the front of the instrument, making sure
they are positioned correctly.

Cause: Prolonged incubation requires continued dispensing of reagent on each slide.
Resolution: For incubation time exceeding one hour, divide protocol step into two,
consecutive steps with the same reagent.

Tissue lifting off slide�

Cause: Tissue section has not properly adhered.

Resolution: Ensure that positively charged slides are used on the VALENT and FFPE
tissue samples are adequately dried in the oven before loading onto the instrument.

Need access to a previous run log to
help determine root cause of weak
staining or failed run�

Cause: The VALENT Software only displays a run log for the most current run, access to
previous run is not available in the VALENT Software.
Resolution: Call Technical Support.
