7. Pre-Start Checklist

Use the Pre-Start Checklist to verify that carboys have been filled, hazardous and non-hazardous carboys have been emptied. Check the
carboy status categories under Bulk & Waste.

'

• Make sure that all reagents and mixing vials have been loaded as shown on the screen and volumes are sufficient. Do not re-use

mixing vials, always use new, clean mixing vials.

• The Bulk & Waste section of Load Reagents window informs the user that carboys have adequate bulk reagent and waste capacity

for the run.

• Check the buffer lines above the Z-head for air bubbles, if air bubbles are present refer to Drain Waste & Prime Buffers of Chapter

5 for instructions on how to remove air bubbles.

• The Delayed Run checkbox on the top right side of the Pre-Start Checklist window allows the user to set a delayed start time for

the run. To set Delayed Run:

o Check the Delayed Run box under Load Reagents
o Select the Start Time
o Select Rinse Slides, and Rinse Buffer Interval Time
o Select Aqua Rinse or Wash Buffer

Important Note: The Delayed Run box only appears once the reagent levels have been checked
• Once everything has been checked, the user can either Start Run or select Delayed Run.
