5.2.2 Microscope Control Panel
Preview Types
In FL Microscope mode the following additional preview modes are available:
◦
Brightfield preview
– a normal type of illumination for scanning. This option is
recommended when scanning in Brightfield mode.
◦
Darkfield preview
– preview image is created with transmitted Darkfield
illumination. The preview is taken from the center of the sample, and the edge of the
slide might be not visible in some cases. Make sure to select this option in Fluorescent
mode.
◦
Pre-scan
– creates a fast FL preview image that is displayed in the Profile Wizard
window. Scanning process takes a couple of minutes only, and it cannot be interrupted.
◦
Pre-scan high-resolution
– creates a High-Resolution fast FL preview image of every
FOV within the selected area (in single layer DAPI, with an illumination time of 20ms) that
is displayed in the Profile Wizard window. Scanning process takes a couple of minutes
only, and cannot be interrupted.
◦
Pre-scan + high-resolution
– creates a High-Resolution fast FL preview image of
every FOV within the selected area (in single layer DAPI, with an illumination time of
20ms) that is displayed in the Profile Wizard window. Scanning process is initiated with a
simple PreScan, the located sample with auto threshold is selected, and finally, an HR
PreScan is launched.
Remove coverslip
If selected, the edges of the coverslip are not detected as specimen to be scanned. Depending on the
threshold level, it is possible that the edges of the coverslip are detected as areas to be digitized.
