5.1.1 Profile Settings Panel

Flash Z-stack
: This function allows the scanning of Z-stack layers with flash illumination. Each
layer is saved as a single slide and are stored in a specific folder. If you use CaseViewer for
viewing slides, please bear in mind that the maximum number of parallel windows that can
be open are 9, therefore it is advised to set layer count to 9 as well.
Step size
determines the spacing between focus levels. One step is around 0.2 µm. Default
settings are:

: 5. The number of iterations the Deconvolution algorithm is
run can be determined in the
Iteration number for deconvolution:
value box.
NOTE:
The purpose of deconvolution is to produce a sharper image, but the use of a
deconvolution algorithm can significantly increase the time that takes to create a digital slide.
The capacity of deconvolution is minimum 96GB RAM.
Scan Settings
Includes the following two scanning modes:
•
Stop and go
: The slide tray stands still during recording the FOV, then it moves on so that the
scanner can record the next FOV.
•
Flash
: This is a much faster method than of the Stop and go. The slide tray is moved in a
vertical line continuously from bottom to top so that a stack of FOVs is recorded during it
movement. The tray stops only at the end of the recorded column.
Use Color matrix:
Select an applicable preset color profile to be used for Brightfield scanning from
the drop-down menu. Settings according to the profile has effect on the color set, color temperature,
and gamma value of the scanned image. Separate color matrix can be defined and used for each
profile and scanning.
Preset profiles
are the following:
•
Calibrated Linear
: Color scheme is defined by calibration (Gamma value is not included)
•
sRGB standard
: Color scheme is defined by calibration (standard Gamma value -2.2- is
applied)
•
sRGB standard Microscope Like
: Color scheme is defined by calibration (standard Gamma
value / White Point: D50)
•
Legacy
: a Color scheme calibrated for the previous software version is applied
•
Custom:
Color profile set in Service is applicable if needed.
Image settings
You can choose the following predefined image formats (and a subsequent custom option) from the
Image Quality
drop-down list in the
Image Encoding
section of the window:
•
