When no more fluid will flow into the cell processing bag or the fluid level
(Figure 6-15 on page 6-17) is just above the rotating seal, close the slide clamp
on the blue-striped tubing.
The capacity of the cell processing bag is approximately 630 mL. If the volume
of the starting cell product is more than 630 mL, not all of the starting cell
product will flow into the cell processing bag during the loading sequence.
Additional starting cell product may be added during the Agitate/Wash-In
operation of the first cycle by doing the following:
a Temporarily clamp the tubing in the valve selected to open during the
first Agitate/Wash-In operation.
b Open the slide clamp on the blue-striped tubing and allow the remaining
starting cell product to flow by gravity into the cell processing bag during
the Agitate/Wash-In operation.
c When all of the starting cell product has flowed into the cell processing
bag, remove the clamp to allow processing/wash solution to fill the cell
processing bag.
d Close the slide clamp on the blue-striped tubing.

Reset the CENTRIFUGE SPEED control to the appropriate setting,
according to the protocol.

Proceed to “Processing” on page 6-26.
Caution:
Verify that fluid is
visible above the
rotating seal when the
centrifuge is spinning to
ensure adequate seal
lubrication. Inadequate
rotating seal lubrication
can lead to failure of the
rotating seal and potential
contamination or loss of
cell product.
