Introduction
Processing Overview
Most cell processing procedures on the COBE 2991 follow the same basic steps.
First, a cell processing set is installed. Next, the cell product bag and
processing/wash solution containers are connected to the cell processing set. The
starting cell product is prediluted (if necessary) and the cell processing bag is filled.
The cell product is then processed either automatically or manually according to the
desired protocol. After processing, the cell product bag is sealed and removed from
the COBE 2991. Finally, the cell processing set, including any waste fluids, is
removed and discarded according to standard operating procedures.
The remainder of this chapter provides an overview of a basic cell processing
procedure, using a 2-liter Wash Packed Red Blood Cells (RBC) protocol as an
example.
Cycles and Operations
Cell processing typically consists of one or more wash/processing cycles, with each
cycle consisting of three operations: Start/Spin, Supernatant-Out, and
Agitate/Wash-In. In Automatic mode, these cycles are pre-programmed and
proceed in order. In Manual mode, the cycles are controlled by the operator; any
operation can be skipped, or any one operation may be used alone.
Prior to initiating a processing procedure, the starting cell product is loaded and air
is removed using the following “preparatory” operations: Predilute, Blood-In, and
Air-Out.
