Never force the centrifuge bowl cover into place. If it does not rotate easily, the cell processing bag may be caught between the centrifuge bowl and
the cover.

Do not pull on the clear tubing connected to the rotating seal. Doing so may cause the rotating seal to fail during processing.

Verify that fluid is visible above the rotating seal when the centrifuge is spinning to ensure adequate seal lubrication. Inadequate rotating seal
lubrication can lead to failure of the rotating seal and potential contamination or loss of cell product.

Do not press STOP RESET during an Air-Out operation. This could lead to excess pressure in the cell processing bag, which may damage the
rotating seal.

The seal weight should move freely on the guide posts. The contacting surfaces should be clean and free of debris and nicks, but not lubricated in
any way. Each seal weight is matched to a particular COBE 2991; seal weights are not interchangeable.

Do not interfere with the seal weight during processing operations. The rotating seal can be damaged and the cell processing set could fail.

The cell processing set’s rotating seal has been validated for continuous processing for up to 90 minutes. Intermittent or extended processing may
result in rotating seal failure and potential loss of cell product.

To maintain rotating seal integrity, a cell product bag rinse procedure must be completed within 3 minutes.

Avoid letting the cell processing set rest for 3 or more minutes after spinning without spinning it again. This minimizes the risk of the rotating
seal faces fusing together, which may result in rotating seal failure.

The COBE 2991 has been validated only for the cell products and processing/wash solutions recommended in the Gambro BCT-approved
protocols.

The spikes on the set are designed for single insertion only.

Place clamps a minimum of ¼ inch from the edge of the hard component at a tubing junction (that is, the junction manifold or spikes).

At the start of the Supernatant-Out operation, ensure that there is an open fluid pathway from the cell processing bag to allow cell product to be
expressed.

When clamping and unclamping lines during the Supernatant-Out operation, press HOLD first. Make the necessary change and then press
CONTINUE. Failure to press HOLD before clamping or unclamping may increase the risk of rotating seal failure.

For protocols that end with resuspended cells, the Agitate/Wash-In operation must be monitored. If the total volume in the cell processing bag
exceeds 450 mL, it will be impossible to remove the centrifuge bowl cover.

For COBE 2991s with pin-diode program boards, do not adjust the spin timers during a spin operation or they could be damaged. Instead, do
the following:
•
For longer spin times, press HOLD. Then, when you wish to resume the program, press CONTINUE.
•
For shorter spin times, press SUPER OUT to move ahead to the Supernatant-Out operation.

The COBE 2991 contains a 1-amp, 125-volt fast-acting ferrule tube fuse in the 24-volt power supply. This fuse is not replaceable by the operator.
Contact a qualified service representative to have the fuse replaced.

Before attempting to reset a circuit breaker, always unplug the power cord.

Whenever a HELP 1 message occurs, contact a qualified service representative.

Do not use ketones, alcohol, or esters for general cleaning of the COBE 2991, except where noted.

Do not use a bleach solution to clean the red cell detector. Damage to the lens will result, which may affect processing outcomes.

The health care institution is responsible for adequately preparing and indentifying product for return shipment. Local regulations may restrict or
prevent shipment of products contaminated with biohazardous material.

Bone marrow must be filtered to remove bone chips and debris before processing on the COBE 2991 system.
Adverse Effects
There are no known adverse effects.
