9. RET scattergram
After fluorescence flow cytometry analysis, a two-dimensional
distribution of the forward scattered light and lateral
fluorescent light is displayed as a scattergram.
In the scattergram, the X-axis represents the intensity of the
lateral fluorescent light and the Y-axis the intensity of the
forward scattered light.
Scattergram analysis algorythms separate the mature red
blood cells, reticulocytes and platelets from each other and
calculate reticulocyte ratio’s, reticulocyte count, optical red
blood cell count and platelet count.
Here, the scattergram is divided into three RET zones based
on the intensity of the fluorescent light and the ratio of the
reticulocytes in each zone to the total number of reticulocytes
is calculated.
