Wipe the outside of the tube to dry it, and then insert the tube gently down 
through the opening in the sample holder block.

In Fl WinLab, start the TimeDrive application and enter the following 
parameters:  

•

 

Duration: 10 minutes 

•

 

Data interval: 0.1 second 

•

 

Excitation wavelength 302nm 

•

 

Emission wavelength 445nm 

•

 

Slits 5nm/5nm Ex/Em 

NOTE:

 

These parameters are only for using coronene as the sample.  If another 

sample is used, then the excitation and emission wavelengths will have to 
be set accordingly.

Start the TimeDrive by clicking the Green traffic light. 

8.

 

Locate the tube center in the focus crossover point to find the highest 
intensity.  
It is useful to look down into the sample tube.  The fluorescence emission is 
in the visible region, allowing you to to see when the sample is being excited 
optimally. 

NOTE:

 

There are two positions that produce high signals. The first is that which 

correctly locates the center of the sample tube in the crossover point of the 
excitation and emission light beams. This is the desired position. The 
second position is that which causes light to be reflected directly from the 
outer surface of the sample tube. This signal is unfortunately often higher 
than the fluorescence signal. Determine that the position achieved after 
alignment is correct by exchanging the sample tube for one containing only 
solvent. If the signal changes dramatically, then the position is optimized 
for fluorescence. If the signal does not change significantly, then the 
position is optimized for scattered light, requiring further alignment.
