To calculate the melting temperatures for a DNA primer pair:

Main Menu
screen, select
Tools Menu

Calculate Tm
to open the
Tm Calculator
screen:

Enter the required information.
a.
Select
Salt Concentration
, then enter the salt concentration in mM.
b.
Select
Primer Concentration
, then enter the DNA concentration in uM.
c.
Enter each primer sequence.
4.
Select
Calculate
.
The melting temperature for each primer is displayed:
