8.2 Troubleshooting Problems with Autotune
Use Table 20 below to diagnose and solve problems with Autotune.
Symptom Cause
Autotune fails at beginning Chromatograph not set to 50 µL/min. If there is no liquid flow from the
chromatograph to push the calibrant into the ion source, the autotune will fail.
Autotune runs for a while
but then fails Chromatograph not set to 50 µL/min. If the calibrant is pushed to the ion source
more quickly, the loop will run out before the instrument refills it. If it is pushed too
slowly, the electrospray may not be stable.
Autotune runs for a while
but then fails If the system is failing autotune with a message that the spray stability check has
failed, check the liquid connections for leaks.
Autotune runs for a while
but then fails An incorrectly adjusted electrospray needle. Adjust electrospray needle.
Autotune runs for a while
but then fails The flow rate out of the liquid pump is unstable. AXP pumps in particular require
high back pressures to provide a stable flow. When using an AXP pump,
backpressure coils should be installed between the pump and the instrument to
allow the pump to run at a stable flow rate.

flowing down the drain
during fill time Out of calibrant. Replace the calibrant bottle.

during fill time Green PEEK tube in the bottle may be above the liquid level. Adjust the position of
the tubing so it is near the bottom of the bottle. It should not touch the bottom or
it may pick up particulates and clog the tubing.

flowing too slowly during fill
time The clear plastic pressurization line, green PEEK, or calibrant loop may be kinked.
Alternatively, the ferrules on the green PEEK tubing may be overtightened.
