Absorbance Plate Test
Description
Before running the Absorbance Plate Test, you must define the
Absorbance Test Plate parameters. See
Defining the Absorbance
Test Plate Parameters
in this chapter for more information.
Gen5’s
Reader
Diagnostics Utility
(PN 5320201) needs to be
installed
before
you can perform the Absorbance Plate Test in Gen5.
This test uses BioTek’s
Absorbance Test Plate
to confirm the Mechanical
Alignment, Accuracy and Linearity, and Repeatability of the ELx800™. The
Absorbance Plate Test compares the reader’s optical density measurements and
mechanical alignment to NIST-traceable values.
To run the Absorbance Plate Test, you will need BioTek’s
6-Filter
(PN 9000547) or
7-Filter
(PN 7260522) Absorbance Test Plate with its accompanying
Standards
Certificate
.
The 7-filter test plate has a glass filter in location C6, which is used to check the
wavelength accuracy of a monochromator. A “peak wavelength test” is performed
during the Absorbance Plate Test in readers equipped with monochromators.
Although the ELx800 does not have a monochromator, you must enter values for
the peak wavelength test, when defining the 7-filter test plate in Gen5™, KC4™, or
KCjunior™ software. The peak wavelength test will not actually be performed on
the ELx800 during the Absorbance Plate Test.

Mechanical Alignment:
The Test Plate has precisely machined holes to
confirm the mechanical alignment of different microplate readers. The
amount of light that shines through these holes indicates whether the
reader is properly aligned. A reading of more than 0.015 OD for any of the
designated alignment wells indicates that the light is being “clipped” and
the ELx800 may be out of alignment. If the reader fails the alignment test,
review the following possible problems and solutions:
Ensure that the Test Plate is correctly seated in the microplate carrier,
with “A1” in the upper left corner. Check the four alignment holes to
ensure that they are clear of debris.

Technical Support.
