at 40°C in order to maintain the sample at the desired temperature while imaging with an oil immersion
objective. To accept this offset you can press
“USE FACTORY” and

allows searching for the best offset value manually, step by step. See
Figure 22 below and paragraph 7.2 for instructions on how to manually calibrate the Objective Heater
(OBJ-COLLAR ####).

automatically finds the best offset according to your specific
experimental conditions. For details on this calibration procedure please follow the instructions on

Figure 24 shows the warning message that will appear once you have pressed
“Calibrate” if you are working
in Chamber Feedback Mode. This message reminds you that you have to ensure that a Chamber Calibration
has already been completed. Or alternatively, that the factory calibration is appropriate for your laboratory
room temperature (i.e. that, even in Chamber Mode, the sample is close to the Set Point temperature). If
you are unsure it is recommended to re-check the room temperature and decide whether to re-calibrate
the chamber.
