Introduction
To perform a run on the HiSeq X, prepare all reagents for your run and then follow the
software prompts to set up the run. Run setup steps include entering run parameters,
loading and priming reagents, loading the flow cell, and performing a fluidics check.
Visit the HiSeq X specifications page on the Illumina website for information about run
duration and other performance specifications.
Sequencing Consumables
Illumina-supplied consumables are required for sequencing on the HiSeq X. Each kit
contains clustering reagents used on the cBot, and SBS reagents, indexing reagents, and
paired-end reagents used on the HiSeq X.

—Each kit includes consumables for sequencing 2 flow cells.
Consumables are packaged in 2 complete sets, each set supports 1 flow cell.

20-pack kit
—Each kit includes consumables for sequencing 20 flow cells. Consumables
are packaged to support 4 flow cells at a time.

Flow Cell and Reagent Preparation
Before setting up the run,
prepare all reagents for the run
: SBS reagents, indexing reagents,
and paired-end reagents. When prompted by the software during run setup, load all
reagents. There is no need to return to the instrument during the run to load reagents.
Allow the packaged flow cell to sit at room temperature for at least 30 minutes before
removing the foil packaging.
