8.2.5.3 Adding Base
After inoculation, an alkaline like Sodium Bicarbonate or Sodium Hydroxide, may be automatically added to the MCV to maintain pH. This
procedure shows how to connect the Base Bottle to automatically add the base solution to the MCV.
NOTE:
Both the Base and Feed pumps only run in one direction, counterclockwise, to advance flow to the MCV.
1. Connect Base tubing line to the MCV.
• Locate the base tubing line (yellow and blue clips) on the MCV headplate.
• Use the AseptiQuik® S connections attached to the Base dip-tube and base addition line.
• Connect the AseptiQuik® S connectors to an audible click.
• Follow AseptiQuik® connection procedure in Section 8.2.1.2.
2. Tubing install in base pump.
• Load base tubing in the base pump. Install tubing line according to the colors indicated on the
base pump cover.
• Twist the gray lever to open and close the pump head.
TIP:
Make sure tubing in the pump head is centered between the V-groove clamp.
• This illustration shows the base bottle set up to automatically add base to the MCV.
3. Prime Base tubing in Manual mode.
• On the HMI, select the base pump icon. Press Program to run in Manual mode.
• Press the Operator icon.
• Stop the pump immediately before base enters the MCV (approximately 15 to 20 mL).
• Return the base pump to program mode.
8.2.6 Media Exchange Phase
The user selects the volume of media from the MCV for removal and replaces it with fresh media. This phase differs from Media Addition
and Media Removal Phases since it allows the user to partially drain or completely drain the bioreactor in addition to the MCV, and
upon restart minimizes transients in pH, DO, and temperature. The user shall ensure that there is sufficient media addition to refill the
bioreactor and maintain the minimum fill level in the MCV.
