Perform Substrate Sampling (typically around 48 hours or 24 hours minimum).
Section 8.3.1
• Cell growth testing, typically at 24 hours or 48 hours. Select Substrate Sampling.
• Open valve on media return line.
• Manually jog the recirculation pump counterclockwise to reduce the bioreactor media volume. Close
valve on the media return line when the bioreactor has reached the desired level.
• Disconnect the FBR temperature sensor.
• Disconnect the Lynx® connectors (if using).
• Remove the FBR to the biosafety cabinet. Perform substrate sampling.
Reconnect the Lynx® connectors after sampling (if using).
• Reconnect the FBR temperature sensor.

Select Media Exchange phase. Media shall be at the correct temperature before addition, otherwise add
cold media slowly.

• Remove Media Reservoir from connector.
• Connect via AseptiQuik® connections to the new Media Addition bottle.
• Connect via AseptiQuik® connections to the Media Waste bottle.
• Open the valve on the return media line.
• Manually jog pump in reverse to empty the FBR to a desired level.
• Close the valve once the bioreactor has reached the desired level.
• Confirm volume sent to the bioreactor. Conditions to stabilize media will auto-start.
