Instruction Manual

Media Exchange Phase

User Action

System Auto Sequence

User initiates Media Exchange at the HMI.

Phase window pops up for the user to enter the following setpoints:

•  Removal Media Pump Flow Rate (mL/min.)
•  Media Removal Volume (mL)
•  Addition Media Pump Flow Rate (mL/min.)
•  Media Addition Volume (mL)
•  Addition Recirculation Pump Flow Rate (mL/min.)
•  Minimum MCV Volume (mL)
•  Stabilization Delay Duration (min.)

User enters setpoints, and selects the play

button.

Prompts user to confirm Current MCV Volume at the HMI.

User confirms the Current MCV Volume at the

HMI.

Prompts user to confirm when all Manual operations are complete.

User manually performs the following:

• Installs via AseptiQuik® connectors the

fresh Media Addition bottle.

• Installs via AseptiQuik® connectors the

Media Waste bottle.

• Confirms all Manual operations are

complete.

Records loop output controller value of all loops.
Resets phase totalizer.
Stops all equipment module control loops. Air mass flow controller is

set to 20 SCCM to prevent backflow into the gas filter.
Orange and red pinch valves are de-energized.
See separate tables for Case A, B and C:

• MCV Volumes is <2.75L (see Situation A).
• MCV Volume is ≥2.75L and the Media Removal Volume is <FBR

Loop Volume (see Situation B).

• MCV Volume is ≥2.75L and Media Removal Volume is ≥FBR Loop

Volume (see Situation C).

User Action

System Auto Sequence

Case A. MCV Volume is <2.75L

• Sets recirculation pump to Operator mode.
• Prompts user to manually jog recirculation pump in reverse to

empty the bioreactor to the desired level, and to confirm when

complete.

User manually jogs pump in reverse to

empty the bioreactor to the desired level and

acknowledges prompt when complete.

Starts media pump counterclockwise at the Removal Flow Rate

setpoint until Media Pump Totalizer reaches Media Removal Volume

setpoint.
Subtracts the Removal Volume from the Current MCV Volume and

Current Total System Volume.
Sets media pump to Operator mode.
Prompts user to jog media pump to add/remove desired volume

from MCV.

User acknowledges prompt to jog media pump. Resets phase totalizer for media and recirculation pumps.

Energizes the red pinch valve to the media reservoir.
Starts media pump clockwise at the Media Pump Addition Flow Rate

setpoint.
When Current MCV Volume reaches (Minimum MCV Volume plus

Bioreactor Loop Volume), starts recirculation pump clockwise at the

Recirculation Pump Addition Flow Rate until the Recirculation Pump

Totalizer reaches the Bioreactor Loop Volume. When Bioreactor Loop

Volume is reached, ramp recirculation pump to the controller values

recorded over 120 seconds.
Continues to run the media pump at the Media Addition Flow Rate

setpoint until the Media Pump Totalizer reaches the Media Addition

Volume setpoint.
Sets media pump to Operator mode.
Adds the volume from Media Pump Totalizer to the Total System

Volume. Subtracts the Bioreactor Loop Volume from the Total System

Volume to calculate Current MCV Volume.
Prompts user to confirm displayed Current MCV Volume and that

media addition is complete or jog media pump until complete.
