3. The steps below are performed automatically by the system:
• Yellow pinch valve is energized to open to harvest wash.
• Blue pinch valve is energized to open to harvest flush.
• Green pinch valve is de-energized to open to harvest solution.
• 3-way valve for harvest pressure is energized to open to pressure control valve
• Pressure control valve is set to 0 psi.
• Resets Recirculation Pump Totalizer.
• Starts recirculation pump counterclockwise at Final Rinse Flow Rate until totalizer reaches Final Rinse Volume.
4.
User shall acknowledge the Final Rinse Phase completion prompt.
8.5 In Situ Lysis Manual Protocol
Harvest consumables are not used in this protocol. This protocol is used in lieu of harvesting the cell in preparation for lysis. The in situ
lysis manual protocol is briefly described as follows:
• Media is drained from the bioreactor and the MCV and may be discarded or kept.
• Solutions are added to the MCV and recirculated through the bioreactor.
• Cells are lysed by lysis solution.
• The bioreactor is washed with DPBS.
• The bioreactor is washed with High Salt solution (optional).
• A small volume of some salt solution is left in the bioreactor.
Use consumables and items listed in Table 8-4.

8.5.1 Remove Cell Culture Media
1. Remove all media from FBR.
• Open the valve on the media return line.
• Run the recirculation pump clockwise (reverse flow) in Manual mode to drain the bioreactor, Lynx® connector and tubing lines, into the MCV.
2. Sampling (optional).
Pull a media sample from the sample collection port during the recirculation.
3. Remove all media from MCV.
• Redirect media to the original media bottle, if still attached.
• Remove tubing from the pinch valve on the left side of the instrument, and clamp off the waste bottle line to force fluid back into the
media bottle.
• Go to Media Removal Phase, press
.

Stop the pump once the lines are empty.
• Turn off the valve on the media return line.
• Reinstall media removal (red/black labels) and media addition (red/white labels) into red
pinch valve on the left side of the instrument.
• Open the clamp on the waste bottle line.
