CORN I NG ASCENT FBR PD SYSTEM

6. Transition to previous phase.

• When Cell Attachment Time is complete, the recirculation pump returns to the bioreactor DO

controller minimum flow rate.

NOTE:

The time to ramp is set by the Ramp Time setpoint.

• The recirculation pump ramps down to BRX DO Min. setpoint (stepwise ramp, every 5 sec.)

7. Sample media (Optional).

• To monitor attachment kinetics, sample the perfusion media through the media sampling

port.

• As an example, media can be sampled every 15 minutes for the first hour and every 30

minutes for the subsequent 2 hours after the system is inoculated with cells.

• Use the system volume to calculate the total number of cells attached as compared to starting

cell number to determine the percentage of cells that have been depleted from the media.

• Alternatively, the concentration of cells in the system can be used to determine the

attachment kinetics.

Inoculation is now complete. Proceed to add base described in Section 8.2.5.3.

8.2.5.1 MCV Inoculation (Preferred Option)
MCV Inoculation is the preferred method. This section lists the MCV Inoculation steps.

On the prompt, select MCV Inoculation.

Follow instructions on the prompt to connect inoculum bottle to the MCV.
