Prime the bottom mobile phase tubing before connecting it to the column. Pump
20% ethanol upflow at a high velocity into the column using a manual command.
Maintain the flow until the bed support is covered and no more air is released from
the bed support. Start and stop the flow several times. Drain the column until approx-
imately 0.5 cm liquid remains. Repeat until all air is removed.

Fit the top distributor on to the adapter. Flush the adapter with 20% ethanol downflow
using a manual command before fitting the bed support. Make sure that the adapter
and tubing are filled with liquid and free from air. Place the adapter securely on e.g.
a flat surface.

Place the wetted adapter bed support in a new snap ring. Fit the bed support and
snap ring assembly onto the distributor as described in this Operating Instruction.
Make sure that the bed support is i contact with the distrubutor all around the cir-
cumference.

If necessary place the adapter (with the bed support downwards) into a container
of 20% ethanol to prevent air from entering the flow-path.

Mount the adapter on to the column as described in this Operating Instructions.

Other equipment
In addition to the AxiChrom column and media, other equipment that is needed is de-
scribed below.
General equipment
The following equipment is needed for all packing methods:
•
Spirit level for leveling the column
•
Media slurry stirrer, to stir the medium in its container and in the column tube.
•
Appropriate tubing and connectors. For more information about the lengths of tubing
needed, see Appendix D Tubing kits and connections, on page 186.
•
Syringe for air purging of the hydraulic chamber.
Equipment needed for Intelligent
Packing
The following equipment is needed to perform Intelligent Packing:
•
Computer with UNICORN 5.01 or higher including Intelligent Packing strategy
•
Compatible ÄKTA system
