High-level overview of isolation protocol with CTS

1.
Inflate and prime the isolation bag.
a.
Add air to the isolation bag to inflate it and enable mixing.
b.
Prime the isolation bag to avoid cells/beads from sticking to the bag.
2.
CTS
™

bead injection and wash.
a.
Wash CTS
™
Dynabeads
™
CD3/CD28 out of the bead input bag and transfer to the isolation
bag.
b.
Wash the beads, remove the supernatant, and transfer to the supernatant bag.
3.
Inject and incubate T cells.
a.
Transfer T cells to the isolation bag.
b.
Incubate for 30 minutes for T cells and beads to bind.
c.
Transfer the supernatant with unbound non-target cells to the supernatant bag.
4.
Wash T cells.
a.
Wash 3X to increase the purity of the isolated cells.
b.
Transfer the wash supernatant to the supernatant bag.
5.
Elute T cells and beads to output.
a.
Elute bead-bound T cells in CTS

Engage magnet, and transfer cell/bead input through the bead removal bag.
