Protocol parameters
The CTS
™
DynaCellect
™
Magnetic Separation System is designed to operate within specific parameters.
The values and limits for each parameter are listed below. If your input value exceeds the maximum
value, an error is displayed.
Parameters that can be modified include:
• Magnet (engaged/disengaged)
• Pinch valves (open/closed)
• Pump speed (range 10–150 mL/min)
• Pump (specified volume or duration)
• Rocker angle (-30°–30°)
• Rocker speed (range 1–30 CPM)
• Bubble sensor (stop when dry/ stop when wet/ inactive)
• Fluid path
Create protocols
Protocols comprise a series of individual steps that are to be performed by the instrument.
Note:
At a high level, there are three parameters that can be set in a step, which are: magnet, rocker,
and pump. The setting of these should be considered for each step.
An isolation and bead removal protocol comprises numerous steps, and the system allows users to
create protocols exactly like they want to. Keep in mind that what may appear as a single step at the
high level may need to be broken down into substeps. For example, “drain isolation bag” can be broken
down into the following steps:
1. Tilt rocker to positive 30 degrees to move liquid towards the sample port,
2. Pump liquid for a certain duration at a certain flow rate to the output bag.
Another example of how a high-level step must be broken down into multiple steps is “fill isolation bag”.
It is advised to keep the isolation bag at a negative angle when filling to avoid foam. Thus, the step
must be broken down into:
1. Move the rocker to negative 30 degrees.
2. Transfer a certain volume from the input bag at a certain flow rate/for a certain duration to the
isolation bag.
