Protein Determination
AOAC method 991.43 suggests that the Kjeldahl method be used for determining
protein content.
Since a large amount of acid is used in the Kjeldahl digestion process, make sure
you use enough base in the Kjeldahl distillation process.
IDF – Kjeldahl Method
To minimize the amount of acid needed for the Kjeldahl method, we recommend that you remove the clear polypropylene
material from the filter bag and run only the filter through the Kjeldahl instrument.
To determine the protein content within the IDF residue, follow the procedure below.
1. Place a clean, blank sheet of paper down on the bench.
2. Working on top of the clean sheet of paper, cut the filter bag just above the seal at the top of the filter making sure
that any loose residue from the bag is collected on the blank sheet of paper.
3. Cut the clear polypropylene skirt in the center from the bottom up to the seal at the top of the filter. Do NOT cut
the white filter material.
4. Peel the clear polypropylene film from the center out to both sides.
