Replace the Mobile Phase
Note:
• It is possible to automatically specify the flow rate and concentration of mobile phase and the oven temperature
after analysis finishes. Refer to
Configure the System to Shut Down after Analysis
• For information about storing a used column, refer to the instruction guide provided with the column or
contact the manufacturer.
• When replacing the mobile phase with a new mobile phase, or when mixing mobile phases, it might be
necessary to replace with another solvent first, to prevent precipitation and emulsion. Refer to
when Replacing the Mobile Phase
Table 5-1 Examples of Solvent Combinations That Should Not be Replaced or Mixed
Directly
Recommended Action
Possible Problem
Solvent Combination
Replace the solvent with
isopropanol.
Emulsion and separation
Water and mobile phase with a low
dielectric constant (hexane,
chloroform, and so on)
Replace the solvent with
HPLC-grade water.
Precipitation
Buffer solution and organic solvent
(methanol, acetonitrile,
tetrahydrofuran, and so on)
Replace the solvent with
HPLC-grade water.
Reaction
Nitric acid and alcohol
1. Press
Pump
to stop pumping.
2. Put the suction tubing for the mobile phase into a reservoir bottle with column sealing liquid and then set the
bottle on the reservoir tray.
3. Purge the mobile phase port to which the suction tubing in the previous step is connected. Refer to
.
4. Configure the pump settings.
