Antibody / Probe Incubation Time
a. The default antibody incubation time is 30 minutes.
b. ISH probe incubation times range from one to four hours, depending on the Template chosen.
Antibody / Probe Incubation Temperature
a. The standard antibody incubation temperature is 25°C (ambient).
b. The maximum antibody incubation temperature is 37°C to avoid reagent evaporation.
c. Probe hybridization temperatures vary, depending on the template chosen.
d. CISH RNA do not require a denaturation step, and typically hybridize at 55°C.
e. CISH DNA protocols include a 95°C denaturation step, typically followed by a 40°C hybridization step.

After making changes to protocols, the user must then click the ‘Save’ button located at the bottom of the ‘Protocol Editor’
window to save the changes.

To delete a protocol a user must first select the protocol by clicking the column to the far left and then select the ‘Delete’ button.
Upon clicking ‘Delete’, the software will ask for confirmation before permanently deleting the protocol.

Click on 'Go Back to Manufacturer Default' to revert to Manufacturer Default Protocols. The sofware will ask for confimation.

To revert a protocol back to the last instance where the ‘Save All Customizations’ button in the ‘Settings’ tab was utilized, select
the protocol to be modified by clicking in the column to the far left and then click the ‘Refresh Protocols with customizations’ button.

Click 'Display current protocol version' to display current version of protocol.

Changes that have not been “saved” may be cancelled by clicking the ‘Cancel’ button.

To close Protocol Editor, click the ‘Return’ button.
