Noise Gating of Dot Plots
The primary purpose of the noise gate is to reduce the saved file size (by eliminating the
quantity of saved events) and correspondingly increase the storage capacity of the disk in terms
of number of tests and images that can be saved.
NOTE: Data gated as noise will not be saved with the file, and once saved you will not be able
to reanalyze this population. Be conservative when excluding noise regions.
Immediately upon acquisition completion the results display an area of events that are grayed
out. This is a default noise gate selection, You will want to further refine this gate as the last
step of the analysis. Selecting the
Gate Adjustment Toggle
icon when the fluorescent (red)
gates are enabled disables the fluorescent gates and enables the noise (yellow) gate
functionality. Touch and drag the yellow marker (images below) to define the data points that
will be identified as noise. As an example, the image below/right shows the edited (saved) test
for the noise setting defined in the image below/middle. These points are also not included in
the population statistics (e.g. total counts, and “Upper Population” and “Lower Population” fields)
even if the size gate includes part of the grayed out region.
Store and Recall of Gate Settings
By default, the gates used in the prior analysis will be retained for the next test. The software
also allows the user to store up to five gate settings in memory for easy recall. Gate settings are
saved by selecting the
STO/RCL
icon (arrow in image below/left). Selecting this icon brings up
a list of five memory locations (numbered zero through four, see image below/middle). Memory
locations that are already occupied are displayed in red and empty locations are displayed in
black. Selecting either location will result in that location being overwritten by the new
fluorescent and size gate settings.
Gates can be recalled at any point (immediately after an acquisition or when analyzing a saved
test) by selecting the same
STO/RCL
icon. Saved gates are displayed in green (see image
