Requirements for the Microscope Stand
The DirectFRAP imaging system is permitted for operation only in combination with the following
microscope stand:
Axio Observer.Z1 (431007-9902-000)
The basic laser safety kit for the Axio Observer.Z1 stand (432927-9011-000) is also required.
The manufacturer does not assume liability for any use that is unrelated to the
intended use.
Any use of the DirectFRAP imaging system for laser manipulation in combination with an
Axio Observer stand having an order number other than that indicated in the present manual,
or without the safety kit for DirectFRAP, is not in accordance with the intended use.
In particular, Carl Zeiss Microscopy GmbH cannot guarantee laser safety in such a case.
Due to the use of high-magnification objectives (e.g.
α
Plan-Fluar 100x/1.45 Oil), it is advisable
to use a vibration protection unit Set up the microscope vibration-free on a vibration-damped
table or on an anti-vibration plate.
Long-term experiments require stable ambient temperatures to avoid any artifacts caused by
thermal expansion.
3.3
Basic Microscope Settings
•
Insert one of the FRAP-capable objectives into the nosepiece, as described in the Axio Observer
operating manual (B 46-0111),
and keep the Immersol 518 F
®
immersion oil ready at hand.
Be sure to read the safety data sheet provided with Immersol 518 F
®
immersion oil.
•
Select suitable fluorescence filter sets (see the table on page 48).
•
Insert the selected reflector modules FL P&C into the reflector turret (see the Axio Observer operating
manual).
