Quitting sample control
When sample control operation is finished, users can press
Return
key (4.3e) to quit sam-
ple control function and return to the main menu of photometric measurement.
Measuring operation
After all measuring parameters are set and white corrections are made, press
Start/Stop
key (4.4c) to start measurement. Each time
Start/Stop
key (4.4c) is pressed, an Abs value is
obtained and the concentration of the measured sample will be calculated. Both the mea-
sured Abs value and the calculated concentration value will be saved in the control unit
and displayed sequentially in the chart on screen. The sample number will increase by 1
after each measurement. The memory of the control unit can store up to 500 groups of
measured data.
The measuring process and the measured results may vary with the different settings
in section
Sample control
(page 51):
- (1) When Reagent White Correction in Cell No. 1 is set to "NO", the system will start
measurement to the set N samples (see
Setting sample cell number in use -Drive Cell No.-
page 51). Then the measured value will be calculated, saved and displayed on LCD one
by one.
- (2) When Reagent White Correction in Cell No. 1 (see
No. 1 Cell white correction
, page 52)
is set to "YES", the system will first measure the set N samples (see
Setting sample cell num-
ber in use -Drive Cell No.-
page 51). Then the measured values of samples No.2~No.N will be
deducted (corrected) by that of the white in sample cell No. 1 following the formula below.
The corrected results will be recorded, calculated and displayed on LCD one by one.
Looking up measured results
During measurement, users can look up the measured results line by line, by pressing
the cursor keys (
,
).
Printing
During measurement, users can print saved measured results when
(F4)
key is pressed.
Quitting quantitative measurement
After finishing quantitative measurement, press
Return
key (4.3e) to quit quantitative
measurement operation. When
Return
key (4.3e)key is pressed, the system will show that
measured data will be deleted. Press
Enter
key (4.3d) to return to the main menu of the
instrument.
Formula of deduction (correction):
Absorbance of sample
n
= measured value of
n
- measured value of sample nº1(white)
Transmittance of sample
n
= measured value of
n
/ measured value of sample nº1(white) × 100%

Users can select a working mode by pressing the corresponding numeric key
1-5
.
Note:
You should wait 15~30 minutes for the instrument to warm up before start wor-
king.
Verifying the installation
After finishing the installation, check sections explained below. You should periodically
check the spectrophotometer's precision and accuracy as well as its base line. To do this,
you should switch the equipment on 30 minutes before doing the test.
Wavelength accuracy and repeatability
The wavelength accuracy should be within ±0.5nm. The wavelength repeatability
should be within 0.2nm. Check with the two spectral lines of the D2 lamp according to the
following procedure:
Confirm the spectral bandwidth is set to 2nm. The initialized spectral bandwidth is
2nm. For more information about setting the spectral bandwidth see
Utilities
(page 70).
Press
(2)
key to select mode
Spectrum measurement
on the main menu.
In Abs or %T mode, press
GoTo WL
(4b) to set the wavelength to 330nm, to turn to the
D2 lamp working mode.
Press (
F1)
key to select parameter setting function. Set the following parameters:
Sampling interval: 0.2nm; Scanning speed: middle; Wavelength range: 660~480nm;
Recording range: 0~100; Mode: Ess.
Press
Start/Stop
(4c) key and set the energy gain value to 5~7 to start scanning.
Press
(F2)
key and set the min. peak/valley value to 1~10, to perform the peak-picking.
Press
(F4)
to print.
Repeat scanning for three times and perform peak-picking and printing. Compare the
picked wavelengths with the standard wavelengths of the two peaks of D2 lamp: 656.1nm
and 486.0nm.
Baseline flatness
With air in sample cell, follow the procedure below to measure the Abs value at whole
wavelength range in spectrum mode:
Confirm that spectral bandwidth is set to 2nm. The initialized spectral bandwidth is
2nm. For more information about setting the spectral bandwidth see
Utilities
(page 70).
Press
(2)
key to select mode
Spectrum measurement
on the main menu.
Press
(F1)
key to select parameter set function. Set the following parameters: Sampling
interval: 1nm; Scanning speed: middle; Wavelength range: 1100~190nm; Recording range:
-0.1~0.1; Photometric mode: Abs.
Press
Autozero
(4.4a) to start baseline correction.
Press
Start/Stop
(4.4c) to start scanning.
Press
(F4)
key to print the spectrum.
Compare the max. Abs value on the spectrum with the value required for the baseline
flatness (sudden change in Abs value is allowed at the point of light source change).
