to place the fluorescent gate at the top/tail of the noise cluster. Gate placement can also be
faciliated by running control (negative or positive) tests, setting the gates (above or below the
control, respectively), storing the gates locations (see section on storing/recalling gates), and
then recalling the gates for the standard, labeled sample.
Noise Gating of Scatter (Dot) Plots
Touching the red gate toggle marker when the fluorescent (red) markers are currently enabled
correspondingly disables the (red) fluorescence gate markers and enables the (yellow) noise
gate markers.
Note: The noise gate is only available after a test is run, before it is saved. It is
not available for saved data.
The user can then touch and drag the yellow circle (images below)
to define the points that will be identified as noise. These noise points will be grayed out (image
below/middle) and will not be saved with the test.
Note: DATA GATED AS NOISE WILL NOT
BE SAVED WITH YOUR FILE!!
Additionally, data in the noise region will not be included in
the stats (counts, sizes, percentages, etc). For example, referring to the image below/right, note
that the lower bead cluster (~6µm), while in the size region and below the red fluorescence gate
line, is not included in the Lower Population counts (i.e. those counts are all zero). This is
because that cluster was included in the noise region during noise gating (below/middle). The
primary purpose of the noise gate was to reduce the saved file size (by eliminating the quantity
of saved events) to correspondingly increase the storage capacity of the disk in terms of number
of tests and images that can be saved. For almost all tests, it is sufficient to leave this gate,
unadjusted, at the system-specified noise exclusion setting.
