6. Troubleshooting Guide
Problem
Solution
Oocyte moves as the PiezoDrill
is actuated.
1.
Pin the oocyte against the bottom of the
dish or slide. See Holding Pipette
section.
Zona is difficult to penetrate.
1.
If a Narishige manipulator is used,
install the Narishige adapter in place of
the ball joint.
2.
Be sure all manipulator and adapter
connections are tight.
3.
Clamp the pipette holder on the
manipulator as closely as possible to the
PiezoDrill.
4.
Check the pipette shape against the
specification (see Pipette section).
5.
Check the fluid being used in the
pipette. Fluorinert (FC-77) is a good
choice. It should fill the pipette
completely.
6.
Initially, apply some positive pressure to
move the Fluorinert close to the tip of
the pipette.
7.
Check the PiezoDrill settings and adjust
as necessary.
Pipette tears zona instead of
coring.
1.
Reduce frequency.
2.
Increase amplitude.
3.
Apply suction.
4.
Pin oocyte to bottom of dish or slide.
Oolemma is difficult to
penetrate.
