3: Operation
Introduction
The main interface for performing measurements is the DataStation software, which
controls the DeltaMyc though the FluoroHub. This section will explain the basics for
operating the instrument to measure single-point lifetimes and to run a lifetime map-
acquisition on a fluorescence sample.
Before running the system
Prior to commencing a measurement it helps to know a few details about the sample,
such as:
Excitation wavelength (
λ
exc
) and emission wavelength (
λ
em
), obtained from steady
state absorption and emission measurements
An approximate idea of the decay time, either from a previous measurement or lit-
erature.
Starting the software

After checking that the appropriate hardware is
connected, turn on the host computer, Delta-
Hub, the Tango stage-controller, and the Del-
taDiode controller.

Plug the lifetime-module power supply in.

For critical measurements we recommend leav-
ing the hardware on for 30 minutes to stabilize.

Launch the DataStation software by clicking on
the icon on the desktop.
This software should automatically find and initialize any accessories present. If
it does not load, check the connections and retry. After DataStation has loaded,
the
New Measurement
window appears. Each icon displayed in the
New Meas-
urement
window corresponds to an availa-
ble measurement option.

To begin a measurement,
select the appropriate
icon, then click the New
button.
